Precis coenia exhibits a seasonal color polyphenism. Animals reared under long-day conditions develop a pale tan pigmentation on their ventral hind wing (the linea form), whereas animals reared under short-day conditions develop a dark reddish-brown pigmentation (the rosa form). This developmental switch is mediated by a shift in the timing of ecdysteroid secretion at the outset of adult development. In addition, a single recessive gene mimics the effect of short-day conditions and, when homozygous, produces a constitutive rosa phenotype. This article analyzes the pigment composition of the ventral hind wing pattern of the linea and rosa forms. It presents a new thin layer chromatography system that separates ommochromes with high resolution. The linea pigmentation was found to be due to xanthommatin, while the rosa pigmentation is due to a mixture dihydro-xanthommatin and ommatin-D. Induction of the rosa form thus requires the synthesis of a new pigment (ommatin-D is a sulfate of dihydro-xanthommatin) and the stabilization of the reduced form of xanthommatin. Arch. Caterpillars of Manduca sexta use two distinct transport mechanisms for the excretion of dyes. One pump (Type A) has a high affinity for acid (anionic) dyes and occurs in the midgut and medial Malpighian tubules. Acid dyes accumulate rapidly in the lumen of the midgut while the Malpighian tubules appear to play only a minor role in the excretion of these dyes. The other pump (Type B) excretes basic (cationic) dyes and is located primarily in the proximal Malpighian tubules. Evidence is presented that hippuric acid competes with acid dyes for excretion by both midgut and Malpighian tubules. After the final-instar larva purges its gut the ability of the midgut and Malpighian tubules to excrete dyes gradually decreases. Sixty hours after the purge only the Malpighian tubules retain some dye excreting activity.  相似文献

Background  

Bacille Calmette-Guerin (BCG) is currently the only available vaccine against tuberculosis (TB) and comprises a heterogeneous family of sub-strains with genotypic and phenotypic differences. The World Health Organization (WHO) affirms that the characterization of BCG sub-strains, both on genomic and proteomic levels, is crucial for a better comprehension of the vaccine. In addition, these studies can contribute in the development of a more efficient vaccine against TB. Here, we combine two-dimensional electrophoresis (2DE) and mass spectrometry to analyse the proteomic profile of culture filtrate proteins (CFPs) from M. bovis BCG Moreau, the Brazilian vaccine strain, comparing it to that of BCG Pasteur. CFPs are considered of great importance given their dominant immunogenicity and role in pathogenesis, being available for interaction with host cells since early infection.  相似文献

Midgut tissue from Aedes aegypti stimulated exflagellation of gametocytes of Plasmodium gallinaceum in the absence of bicarbonate, a factor necessary for in vitro exflagellation. Exflagellation was also stimulated when washed infected red cells in a buffered saline (pH 7.4) not containing bicarbonate were introduced into the midgut by enema. The exflagellation-stimulating activity was neither sex nor species specific. Preparations of a mosquito exflagellation factor (MEF) were obtained without tissue disruption by collecting the fluid excreted by Anopheles stephensi females while they were feeding on warm saline. MEF was dialyzable and stable to boiling and decarbonation. Thus, MEF is not bicarbonate.  相似文献

Background

Dopamine is a catecholamine that is used as a neurotransmitter both in the periphery and in the central nervous system. Dysfunction in various dopaminergic systems is known to be associated with various disorders, including schizophrenia, Parkinson's disease, and Tourette's syndrome. Furthermore, microdialysis studies have shown that addictive drugs increase extracellular dopamine and brain imaging has shown a correlation between euphoria and psycho-stimulant-induced increases in extracellular dopamine [1]. These consequences of dopamine dysfunction indicate the importance of maintaining dopamine functionality through homeostatic mechanisms that have been attributed to the delicate balance between synthesis, storage, release, metabolism, and reuptake.

Methods

We construct a mathematical model of dopamine synthesis, release, and reuptake and use it to study homeostasis in single dopaminergic neuron terminals. We investigate the substrate inhibition of tyrosine hydroxylase by tyrosine, the consequences of the rapid uptake of extracellular dopamine by the dopamine transporters, and the effects of the autoreceoptors on dopaminergic function. The main focus is to understand the regulation and control of synthesis and release and to explicate and interpret experimental findings.

Results

We show that the substrate inhibition of tyrosine hydroxylase by tyrosine stabilizes cytosolic and vesicular dopamine against changes in tyrosine availability due to meals. We find that the autoreceptors dampen the fluctuations in extracellular dopamine caused by changes in tyrosine hydroxylase expression and changes in the rate of firing. We show that short bursts of action potentials create significant dopamine signals against the background of tonic firing. We explain the observed time courses of extracellular dopamine responses to stimulation in wild type mice and mice that have genetically altered dopamine transporter densities and the observed half-lives of extracellular dopamine under various treatment protocols.

Conclusion

Dopaminergic systems must respond robustly to important biological signals such as bursts, while at the same time maintaining homeostasis in the face of normal biological fluctuations in inputs, expression levels, and firing rates. This is accomplished through the cooperative effect of many different homeostatic mechanisms including special properties of tyrosine hydroxylase, the dopamine transporters, and the dopamine autoreceptors.  相似文献

Polyphenisms are evolved adaptations in which a genome produces discrete alternative phenotypes in different environments. In this study, the genetic basis of the evolution of a polyphenism by genetic accommodation was investigated. A polyphenic strain and a monophenic strain of Manduca sexta (L.) were crossed and the F(1) offspring and backcross progeny were analysed. The larval colour polyphenism was found to be regulated by one sex-linked gene of major effect and many smaller effect modifier genes. The finding shows that the mechanism of genetic accommodation relies on genetic changes that are consistent with the current view of the genetic basis of adaptive evolution.  相似文献

Complex Regional Pain Syndrome type one (CRPS I) or formerly Reflex Sympathetic Dystrophy (RSD) is a disabling syndrome, in which a painful limb is accompanied by varying symptoms. Neuropathic pain is a prominent feature of CRPS I, and is often refractory to treatment. Since gabapentin is an anticonvulsant with a proven analgesic effect in various neuropathic pain syndromes, we sought to study the efficacy of the anticonvulsant gabapentin as treatment for pain in patients with CRPS I.  相似文献

Morphologically distinct worker castes of eusocial insects specialize in different tasks. The relative proportions of these castes and their body sizes represent the demography of a colony that is predicted to vary adaptively with environments. Despite strong theoretical foundations, there has been little empirical evidence for the evolution of colony demography in nature. We show that geographically distinct populations of the ant Pheidole morrisi differ in worker caste ratios and worker body sizes in a manner consistent with microevolutionary divergence. We further show that the developmental mechanism for caste determination accounts for the unique pattern of covariation observed in these two traits. Behavioral data reveal that the frequency of different tasks performed by workers changes in a caste-specific manner when caste ratios are altered and demonstrate the importance of the major caste in colony defense. The population-level variation documented here for P. morrisi colonies supports the predictions of adaptive demography theory and illustrates that developmental mechanisms can play a significant role in shaping the evolution of phenotype at the colony level.  相似文献
